Mode Of Action

Inhibition of DNA-dependent RNA polymerase activity in susceptible bacterial cells.

Drug Indication Pulmonary TB (with other drugs); prophylaxis of Mycobacteria avium complex infection in immunocompromised patients with low CD4 count; non-tuberculous mycobacterial infections (with other drugs).
Precautions To be administered always with another tuberculostatic agent, pregnancy, impaired liver functions, undernourished patients, late pregnancy (can cause post-natal haemorrhage), immunologic reactions with intermittent therapy, discontinue immediately if signs of hepatocells are noted, porphyria
Contra-Indications Jaundice, impaired liver functions, lactation and pregnancy, new born infants.
Side Effects Elevated liver enzymes; jaundice; hepatitis (not common); nausea and vomiting; anemia; leucopenia; cytopenia; haemolysis (not common); hypersensitivity reactions; coneal opacities (with long term use); orange-red colouration of urine, saliva and skin; arthralgia and myalgia; flu-like syndrome.
Dosage Pulmonary TB (with other drugs): 150-450mg OD for upto 6 months after culture negative. Prophylaxis of Mycobacteria avium complex infection in immunocompromised patients with low CD4 count: 300mg OD; non-tuberculous mycobacterial infections (with other drugs): 150-600mg OD for upto 6 months after culture negative.
